“氧感知通路”于2019年获诺贝尔生理学或医学奖,作为一种关键的调控蛋白,低氧诱导因子-1(hypoxia inducible factor-1,HIF-1)受到广泛关注,其普遍存在于人和哺乳动物细胞内,有相当广泛的靶基因谱,其中包括与缺氧适应、炎症发展及肿瘤生长等相关的近100种靶基因。

低氧诱导因子(HIF-1)在人类的代谢调节中,属于一个核心角色。在细胞中,HIF 信号级联反应会受到缺氧状态的影响。在缺氧状态下,通常会让细胞持续的细胞分化。然而,缺氧状态促进了血管新生,对于胚胎中的血管系统与癌症肿瘤来说非常重要。伤口处的缺氧状态,也促进了角质细胞的移动与上皮组织的修护。在普遍情况下,HIF 也是发育的重要关键。在哺乳动物中,若缺少了HIF-1的基因,将导致胎儿死亡。HIF-1已经被证实,对于软骨细胞的存亡有重大的影响,它能使软骨细胞适应在骨骼间生长板的缺氧环境。

揭示生物氧气感知通路,不仅在基础科学上有其价值,还有望带来创新的疗法。比如倘若能通过调控HIF-1通路,促进红细胞的生成,就有望治疗贫血。而干扰HIF-1的降解,则能促进血管生成,治疗循环不良。另一方面,由于肿瘤的生成离不开新生血管,如果能降解HIF-1α或相关蛋白(如HIF-2α),就有望对抗恶性肿瘤。
